SITC 2026: Modular Lipid Bilayer Platform Enables Antigen-Specific vs. Polyclonal CD19 CAR-T Cell Expansion with Distinct Phenotypic and Functional Outcomes

A single programmable RBC platform supports feeder-free T-cell activation and antigen-specific CAR-T cell expansion, with quantitatively controlled antigen presentation. sbPCI CD3/CD28 produced more T cells at electroporation and more CAR+ cell than competitive nanomatrix. sbPCI CD19/28/137 gave higher CAR+ expansion and moved toward a more balanced CD4:CD8.

SITC 2026: Synapse-localized Gamma-chain Cytokine Presentation Enables Single-dose, Soluble Cytokine-free T cell Expansion in Plate and G-Rex with Reduced Manufacturing Complexity and Cytokine Dosing

Integrated cytokines in this formulation support expansion with reduced metabolic demand while favoring memory associated T cell phenotypes. Synapse-localized cytokine presentation, through surface integration via sbPCI, supported effective T cell expansion in 6M G-Rex while favorably shifting differentiation toward memory-associated, cytotoxically armed phenotypes.

Allison Symposium 2026: Spatial Control as a Third Axis of Immune Regulation: Signal Placement Acts Independently of Receptor Identity and Signal Strength

Immune cells respond not only to what signals they receive and how much they receive, but also to how those signals are spatially organized. Signal arrangement influences immune cell phenotype and function independent of receptor identity and signal strength. They product distinct T cell and NK cell responses when presented in different spatial configurations. This indicates that signal geometry represents a programmable and previously underexplored determinant of immune cell outcome.

CellPilot™ Explorer: Engineer Cell Lines with Evidence, Not Assumptions

For immune cell programs, the bottleneck is not only biology. Cell line development often requires engineering that takes up to 9 months of cloning and qualification. Add on cell line maintenance and you burn through significant material and budget before you can screen for the best option. CellPilot Explorer reduces the time and cost of configuring and optimizing signal design.

CellPilot™ Custom: Custom differentiation in weeks, without the workload

When standard differentiation protocols can’t achieve the desired outcome, identifying the optimal combination of biological signals can take months of iterative testing. Challenging cell types, specialized phenotypes, and emerging therapeutic applications often require signaling approaches that are not addressed by off-the-shelf solutions. CellPilot Custom accelerates discovery and optimization of tailored signal combinations.

ASGCT 2026: Rapid, Feeder-Free Expansion of NK Cells in Less Than 14 Days Using a Biocompatible Lipid Bilayer-Based Immunomodulating Expander in a Bioreactor System

sbPCI-NK Expander enables feeder-free NK expansion from unfractionated PBMCs without NK-specific media. sbPCI-IL21 activates STAT3 equivalently to soluble delivery yet produces distinct receptor and cytotoxic profiles, indicating that immune synapse architecture, not signal strength alone, is a tunable lever for NK manufacturing.

Speed Meets Precision: Tailored Immune Cell Function through Rapid Quantitative Immunology

StimulusBio’s Programmable Cellular Interface (sbPCI) technology delivers a transformative solution for immune cell screening and functional characterization. The technology enables direct functionalization of cells by enabling rapid, modular presentation of bioactive molecules (antigens, costimulatory ligands, cytokines, etc.) on biocompatible presenting cells.

Accelerating Cell Therapy: Navigating Immune Mediation with Comprehensive Cell Interaction Screening

StimulusBio’s CellPilot platform charts a faster, more cost-effective course for screening immune cell interactions. Designed to guide scientists through complex cellular landscapes, CellPilot acts as a navigation system for immune cell discovery. Powered by sbPCI technology, CellPilot enables a broader interrogation of cellular mediators. This streamlined approach eliminates the need for time-consuming genetic modification and cuts months off traditional timelines.

SITC 2025: A Configurable, Cross-Modality Extracellular Vesicle Standard (EV-CMS) Enables Quantitative Evaluation Across Nanotechnology Platforms

EV-CMS, based on sbPCI technology, has potential to enable more accurate EV detection and control. sbPCI enables quantitative labeling of lipid bilayers, including extracellular vesicles. The incorporation of stoichiometrically-controlled labels that can be detected across distinct modalities enable cross-platform quantitation of extracellular vesicles, with the same EV-CMS.

SITC 2025: Selective Programming Drives Faster Expansion of Antigen-Specific T cells to Improve Cell Therapy Manufacturing and Outcomes

sbPCI Eliminates Isolation Steps and Enables Rapid Expansion of Rare TCR-specific Cells to Clinically Sufficient Numbers in 8 Days. The technology can be successfully customized to product clinically-relevant numbers of TCR-specific T cells from a large polyclonal population within 8 days. Using sbPCI, CMV TCR-specific CD8+ T cells were selectively expanded from a starting population with low CMV-TCR expression levels. After 48 hours, the expanded T cells produced cytolytic and pro-inflammatory cytokines.

SITC 2025: High Throughput Screening of a Programmable Cellular Interface (sbPCI) Rapidly Identifies Configurations Capable of Expanding T Cell Subsets Without Isolation

sbPCI enables high-throughput, cost-effective identification of donor-specific formulations for T-cell expansion that outperform industry standards. The technology simplified and accelerated screening with assay setup reduced to 2-4 hours and scale increased to >500 samples/assay. The assay identified CD3, CD28 and CD137 configurations combined with IL-2 that better enabled T cell expansion compared to industry standard. Phenotype-specific formulations were also identified.

SITC 2024: Development of Corticosteroid-Resistant T-cell Therapy for Transplant and Autoimmune Disease Diagnosed Patients.

Dexamethasone resistant cell therapy is achievable and scalable for patients undergoing long-term immunosuppressive treatment. The treated cells expressed similar levels of cytolytic enzymes as untreated cultures and expanded a majority of CD3+CD8+ cells. The results support the viability of manufacturing potent corticosteroid-resistant therapies for patients.
